Within STROKEMARK, the research line on 2D and 3D cell models explores the role of peripheral blood mononuclear cells (PBMCs) and their subtypes (CD34+/CD14+, CD34−/CD14−, etc.) in ischemic stroke. Cells derived from patients are analyzed for gene and protein expression (p66Shc, JunD, Sirt5) using 2D cultures, while in vivo validation is based on experimental mouse stroke models (wild-type, transgenic, and knockout mice). Functional tests include angiogenic and reparative capacity, oxidative stress regulation, and impact on blood-brain barrier integrity. This approach allows the integration of cell biology with clinical and biomarker data, clarifying the interaction between aging genes, inflammation, and stroke prognosis.

Cell models represent a translational bridge between molecular mechanisms and clinical outcomes in patients. Through the characterization of PBMC subtypes and their functional responses, this line of research can identify circulating cellular signatures predictive of recovery or post-stroke complications. These models can be used to test pharmacological interventions targeting inflammatory or aging pathways in controlled environments prior to their clinical application. 3D and in vivo systems allow the validation of new therapeutic targets, providing platforms for preclinical drug screening. Furthermore, the integration of patient-derived samples with animal models supports precision medicine strategies, promoting personalized rehabilitation and secondary prevention programs. In the long term, cellular models could foster innovation in regenerative therapies and immune modulation in cerebrovascular diseases.
